Описание:

In recent years, a tremendous amount of effort has been focused on better understanding the fundamentals of Alzheimer's disease (AD) to facilitate early and accurate diagnosis and appropriately targeted therapeutic treatments. Alzheimer's Disease: Targets for New Clinical, Diagnostic, and Therapeutic Strategies provides a detailed synopsis of the current state of the art of diagnostics and therapeutics and identifies emerging technologies and molecules that show promise in the management and treatment of AD.

With contributions from experts drawn from academia, clinical practice, and the biotechnology and pharmaceutical industries, the book explores:

  • The basis of AD and the role of Aβ oligomers in development of disease
  • Existing and emerging in vitro biomarker-based methodologies for the diagnosis of AD, focusing on genetic, biochemical, and conformational strategies
  • In vivo imaging diagnostic approaches
  • Evolving diagnostic criteria, health regulatory guidelines, biomarkers in clinical trials, and available and emerging therapies
  • Recent progress in small-molecule disease-modifier drug discovery efforts for AD, specifically in the areas of Aβ, tau, and emerging neuroprotective/neurorepair approaches
  • How a case study of AD raises issues regarding clinical and pathologic criteria, risk factors, and the amyloid hypothesis
  • The molecular conformational factors that govern the pathogenicity of aggregating proteins, and how these factors could represent new targets for disease-modifying therapies
  • The latest epidemiological, pathological, biochemical, and behavioral studies that may shed some light on the risk of developing AD and similar dementias after traumatic brain injury

Examining current hypotheses and suggesting possible new approaches to therapeutic clinical applications, this volume paves the way for a robust pipeline of therapeutics to combat not only AD, but a whole host of other neurodegenerative diseases.

Описание: Parkinson’s Disease: Molecular Mechanisms Underlying Pathology explores the molecular pathways at the basis of the cellular defects connected to Parkinson’s disease, the second most common neurodegenerative disease, and the most common movement disorder. This book presents the latest research on the pathways and mechanisms that have been discovered to play a role in Parkinson's pathology. . This focus on mechanisms rather than individual genes allows the contributors to elaborate on overlapping and joint functions of different causative genes.Readers will find descriptions of model systems that present parallels (and differences) between discoveries in different species, demonstrating the importance of multidisciplinary research that spans a broad array of technologies and model organisms. Written from both a cross-methodology and cross-species perspective, the book provides readers with the current state of knowledge on the molecular biology of Parkinson’s.

Описание: The number of patients with dementia is rising in proportion to the aging of our population; therefore, by 2050 the prevalence of Alzheimer's disease is expected to triple, risking the integrity of the entire U.S. healthcare economy. More NIH dollars are spent on Alzheimer's disease research
than on any other neurological disease and our available fund of information has commensurately accelerated. Whereas 30 years ago little distinction was made between different forms of dementia, clinicians now distinguish multiple degenerative and vascular syndromes, each with their own etiologic,
therapeutic and prognostic implications. New genetic insights reach the popular press on a regular basis. Consequently Alzheimer's disease (and related disorders) has become a special concern to the majority of the population whose main avenue of care lies with the primary care physician.

The primary care physician (general internist, family practitioner, nurse practitioner), geriatrician, and geriatric psychiatrist are the target audiences for this book; in short anyone who is not a neurological expert who provides dementia-related care. As the primary care physician is the target
audience for this title, the volume begins with a discusion of how to produce an effective diagnosis through a series of precise, easy- to- follow steps. Additional sections detail the current research state of each of the major individual dementia diseases that the general clinician frequently
encounters. The last section procides a clinically focused 'how to' guide for treatment including symptom-directed pharmacotherapy, as well as an up-to-date summary of ongoing major therapeutic trials and an assessment non-pharmacological management issues such as driving and end-of- life
considerations. Aiming to be clear, consice and practical, this volume provides the essential information in dementia related care, from disease diagnosis to symptom management. Because it uses the most up-to-date dementia research available, this volume will become a close companion of the
primary care physician, who seeks direct answers to the common questions and issues that arise in the diagnosis and management of patients with dementia.

Описание: This volume brings together various theories of how aberrations in mitochondrial function and morphology contribute to neurodegeneration in idiopathic and familial forms of Parkinson’s disease. Moreover, it comprehensively reviews the current search for therapies, and proposes how molecules are involved in specific functions as attractive therapeutic targets. It is expected to facilitate critical thought and discussion about the fundamental aspects of neurodegeneration in Parkinson’s disease and foster the development of therapeutic strategies among researchers and graduate students. Theories of idiopathic Parkinson’s etiology support roles for chronic inflammation and exposure to heavy metals or pesticides. Interestingly, as this project proposes, a case can be made that abnormalities in mitochondrial morphology and function are at the core of each of these theories. In fact, the most common approach to the generation of animal and cell-culture models of idiopathic Parkinson’s disease involves exposure to mitochondrial toxins. Even more compelling is the fact that most familial patients harbor genetic mutations that cause disruptions in normal mitochondrial morphology and function. While there remains to be no effective treatment for Parkinson’s disease, efforts to postpone, prevent and “cure” onset mitochondrial aberrations and neurodegeneration associated with Parkinson’s disease in various models are encouraging. While only about ten percent of Parkinson’s patients inherit disease-causing mutations, discovering common mechanisms by which familial forms of Parkinson’s disease manifest will likely shed light on the pathophysiology of the more common idiopathic form and provide insight to the general process of neurodegeneration, thus revealing therapeutic targets that will become more and more accessible as technology improves.
