Описание: Specific biomolecular interactions occur in practically every bio- logical process, usually involving macromolecules such as proteins and nucleic acids. Thus, it is not surprising that great attention has been devoted by biologists, chemists, and physical chemists to the general phenomenon of macromolecular recognition in order to gain a better understanding of the principles, forces, and energetics that de- termine the often remarkable specificities of such interactions as those of enzymes and their substrates and inhibitors, antibodies with anti- gens, hormones with their receptors, and DNA with drugs. These studies have allowed the unraveling of some basic principles of mol- ecular recognition, such as complementarity, specificity, and dynam- ics of the interacting species, and at the same time have led to development of analytical and separation techniques based on bio- logical specificity (e. g., affinity chromatography and other bioaffinity methods). This volume was assembled to address current research and developing ideas in biomolecular recognition through the contribu- tions presented at the Conference on Mechanisms of Recognition in Biological Macromolecules held in Siena, Italy, September 4-6,1986. The Conference was cosponsored by the National Research Council of Italy, the Italian Biochemical Society, the University of Siena, and the International Interest Group in Biorecognition Technology.

Описание: Biological processes involving large macromolecular assemblies are thought to be a dynamic consequence of cooperativity and metastability. The folding of a peptide chain creates local environments from which "activity" can emerge. In the same way, the assembly of large molecular complexes creates dynamic features that would only be feasible in a large construct.

The biological implications of such adaptation will be explored as it applies to the static quasisymmetry situations, as well as to the dynamics of structural transitions. The current wealth of solved high-resolution complex structures makes this an appropriate time to summarize the state of the art in structural dynamics of living architectures. With contributions by leading scientists in the fields of virology, bacterial flagellum, cytoskeleton, ribosome and giant enzymes, this work presents knowledge in the various fields of structural proteomics of very large molecular assemblies, with focus on their mechanisms of action.

Описание: Throughout most of this century, progress in Polymer Science took place by way of studies of dilute polymer solutions and of the solid state. More recently, attention has been focused on the semi-dilute regime. The elucidation of transi- ent network and temporal gel states under conditions of moderate concentra- tion by numerous hydrodynamic and optical techniques is now well established and a solid theoretical underpinning for these phenomena is in place. However, in most of the systems studied, interpolymer interactions are relatively weak forces, because in solutions oftypical polymers, strong interpolymer interactions lead to bulk phase separation. However, polymers that form highly associating structures in equilibrium solution now constitute an important class of synthetic macromolecules, and it is evident that strong intermacromolecular interactions - without accompanying phase separation - are prevalent in biopolymer sys- tems as well. This behavior is generally characteristic of amphiphilic polymers, which contain both hydrophilic and hydrophobic repeat units. It is also charac- teristic of systems in which two macromolecular species are complementary, such as hydrogen-bond-donating and hydrogen-bond-accepting polymers, or polycations and polyanions. Interest in associating polymer systems currently derives from both applied and fundamental questions. Synthetic water-soluble polymers bearing hydro- phobic groups exhibit special rheological properties that make them well-suited as thickeners and viscosity-modifiers. Drag-reduction effects may arise in such polymer solutions, and may also be observed in polymer complexes based on hydrogen-bonding.
