Описание: Proteinases were among the first enzymes to be investigated biochemi- cally, and purification and crystallization especially of proteolytic enzymes of the digestive tract has contributed much to our present knowledge of enzymic structure and mechanisms of catalysis. However, for a long time little has been known about the functional aspects of proteinases. The only exception from this have been the digestive tract enzymes responsible for extracellular catalysis of protein breakdown and supply of amino acids for new-protein assembly and nitrogen metab- olism in the respective organs. The work of Schoenheimer, summarized for the first time in 1942 in a paper entitled Dynamic state of body constituents, showed that continuous turnover of proteins takes place in cells. But scientists did not pay much attention to these findings at that time. The continuous accumulation of knowledge of a variety of intracellular proteolytic processes during the past decades has greatly stimulated research in this field. The central role of proteo- lysis in cellular regulation has become fully evident during recent years. It is the aim of the 30th Mosbach Colloquium to present an over- view of our present knowledge of proteinase structure, function and control. The relationship between globular protein structure of a proteinase and induction of enzymic activity will be discussed for trypsin and trypsinogen activation. One significant proteinase action is the total degradation of proteins to serve cellular needs under different condi- tions.

In consequence, it is treated in most text- books of biology and biochemistry at least at an elementary level. However, certain areas of carbohydrate biochemistry have expanded rapidly over the past few years as a result of intense research activity. Such advances make this an opportune time to draw together the newer ideas of carbohydrate functions and to present them within a framework of established knowledge. This book describes the role of carbohydrates in biology at a level which is suitable for undergraduate students in biochemistry, biology and the medical sciences. The subject is treated to the final-year level although, inevitably, advanced students will wish to expand their knowledge of particular topics by use of the reading list provided as an appendix. Two areas are particularly emphasized. The first is the metabolic reactions of carbohydrates and the functions of such reactions in the organism. Here the regulation of metabolism in response to different physiological requirements is stressed, and concepts of compartmentation and transport are explored. The second area is concerned with the metabolism and functions of carbohydrate polymers. Here there has been a rapid expansion of research, particularly into the role of specific interactions of carbohydrates which occur, for example, at cell surfaces.

Описание: Since its ?rst description in 1942 in both serum and cerebrospinal ?uid, transthyretin (TTR) has had an eventful history, including changes in name from “prealbumin” to “thyroxine-binding prealbumin” to “transthyretin” as knowledge increased about its functions. TTR is synthesised in a wide range of tissues in humans and other eutherian mammals: the liver, choroid plexus (blood- cerebrospinal ?uid barrier), retinal pigment epithelium of the eye, pancreas, intestine and meninges. However, its sites of synthesis are more restricted in other vertebrates. This implies that the number of tissues synthesising TTR during vertebrate evolution has increased, and raises questions about the selection pressures governing TTR synthesis. TTR is most widely known as a distributor of thyroid hormones. In addition, TTR binds retinol-binding protein, which binds retinol. In this way, TTR is also involved with retinoid distribution. More recently, TTR has been demonstrated to bind a wide variety of endocrine disruptors including drugs, pollutants, industrial compounds, heavy metals, and some naturally occurring plant ?avonoids. These not only interfere with thyroid hormone delivery in the body, but also transport such endocrine disruptors into the brain, where they have the potential to accumulate.
