Описание:

Alzheimers disease (AD) is the leading cause of dementia and, unfortunately, remains incurable. The social, emotional and financial implications of AD are immeasurable, and about 47 million people worldwide are affected by AD or other forms of dementia. As lifespans are improved by healthcare systems worldwide, age-associated neurodegenerative diseases are imposing an increasing challenge to science. It is becoming imperative for us to understand the causes of these diseases, AD in particular, at molecular and cellular levels. Starting with the broader picture from a biological perspective, this book takes the reader through fascinating dynamics within and outside of neurons in the brain.

Alzheimers Disease: Biology, Biophysics and Computational Models helps the reader to understand AD from mechanistic and biochemical perspectives at intra- and inter-cellular levels. It focuses on biochemical pathways and modeling associated with AD. Some of the recent research on biophysics and computational models related to AD are explained using context-driven computational and mathematical modeling and essential biology is discussed to understand the modeling research.

The Latest Developments on the Role of Dynamics in Protein Functions

Computational Approaches to Protein Dynamics: From Quantum to Coarse-Grained Methods presents modern biomolecular computational techniques that address protein flexibility/dynamics at all levels of theory. An international contingent of leading researchers in chemistry, physics, and biology show how these advanced methods provide insights into dynamic aspects of biochemical processes. A particular focus is on intrinsically disordered proteins (IDPs), which lack a well-defined three-dimensional structure and function as dynamic ensembles.

The book covers a wide spectrum of dynamics, from electronic structure-based to coarse-grained techniques via multiscaling at different levels. After an introduction to dynamics and historical overview of basic methodologies, the book addresses the following issues:

  • Is there a quantitative relationship between enzymatic catalysis and protein dynamics?
  • Which are the functionally relevant motions of proteins?
  • How can structural properties and partner recognition mechanisms of IDPs be simulated?
  • How can we speed up molecular dynamics?
  • How can we describe conformational ensembles by the synergistic effort of computations and experiments?

While dynamics is now considered essential for interpreting protein action, it is not yet an integral component in establishing structure-function relationships of proteins. Helping to reshape this classical view in biochemistry, this groundbreaking book explores advances in computational methodology and contributes to the new, ensemble way of studying proteins.

Computational Chemotaxis Models For Neurodegenerative Disease

Описание:

The mathematical model presented in this book, based on partial differential equations (PDEs) describing attractant-repellent chemotaxis, is offered for a quantitative analysis of neurodegenerative disease (ND), e.g., Alzheimer's disease (AD). The model is a representation of basic phenomena (mechanisms) for diffusive transport and biochemical kinetics that provides the spatiotemporal distribution of components which could explain the evolution of ND, and is offered with the intended purpose of providing a small step toward the understanding, and possible treatment of ND.

The format and emphasis of the presentation is based on the following elements:

  • A statement of the PDE system, including initial conditions (ICs), boundary conditions (BCs) and the model parameters.
  • Algorithms for the calculation of numerical solutions of the PDE system with a minimum of mathematical formality.
  • A set of R routines for the calculation of numerical solutions, including a detailed explanation of all of the sections of the code. The R routines can be executed after a straightforward download of R, an open-source scientific computing system available from the Internet.
  • Presentation of the numerical solutions, particularly in graphical (plotted) format to enhance the visualization of the solution.
  • Summary and conclusions concerning the principal results from the model that might serve as the basis for a next step in the modeling of ND.

In other words, a methodology for numerical PDE modeling is presented that is flexible, open ended and readily implemented on modest computers. If the reader is interested in an alternate model, it might possibly be implemented by: (1) modifying and/or extending the current model (for example, by adding terms to the PDEs or adding additional PDEs), or (2) using the reported routines as a prototype for the model of interest.

These suggestions illustrate an important feature of computer-based modeling, that is, the readily available procedure of numerically experimenting with a model. The current model is offered as only a first step toward the resolution of this urgent medical problem.

Описание:

The accessibility of the skin in vivo has resulted in the development of non-invasive methods in the past 40 years that offer accurate measurements of skin properties and structures from microscopic to macroscopic levels. However, the mechanisms involved in these properties are still only partly understood. Similar to many other domains, including biomedical engineering, numerical modeling has appeared as a complementary key actor for improving our knowledge of skin physiology.

This book presents, for the first time, the contributions that focus on scientific computing and numerical modeling to offer a deeper understanding of the mechanisms involved in skin physiology. The book is structured around some skin properties and functions, including optical and biomechanical properties and skin barrier function and homeostasis, with--for each of them--several chapters that describe either biological or physical models at different scales.
