Описание: This book shows how physics can be used to analyze the changes that cells and tissues undergo during development. Major stages and components of the biological development process are introduced and analyzed. Full-color throughout, this comprehensive textbook is suitable for graduate and upper-undergraduate courses in physics and biology.
Описание: This volume covers the current knowledge base on the role of signaling and environmental pathways that control the normal development of germline stem cells, meiotic progression of oocytes, events of oocyte maturation and fertilization, and the birth of an embryo. Germ cells are uniquely poised to sustain life across generations through the fusion of oocyte and sperm. Because of the central importance of germ cells to life, much work has been dedicated to obtaining a clear understanding of the molecular and signaling events that control their formation and maintenance. Germ cells are set aside from somatic cells in the embryo and go through specialized meiotic cell cycles as the animal matures. These cell cycles are interspersed with long periods of arrest. In human females, meiosis I is initiated in the fetus. At birth, oocytes are arrested in meiosis I; after puberty, every month an oocyte initiates meiosis II – ovulation. Upon sperm availability these cells are fertilized, generate an embryo, and the cycle-of-life continues. During meiotic I progression and arrest, the fitness of oocytes and their progeny are likely influenced by environmental cues and signaling pathways. A lot of recent work has focused on understanding the mechanisms that regulate oocyte fitness and quality in humans and vertebrates. Much of our understanding on the events of meiosis I and germline stem cell populations comes from work in invertebrates, wherein the germline stem cells produce oocytes continuously through adult development. In both inverbrates and vertebrates nutritional and signaling pathways control the regulation of stem cells in such a manner so as to couple production of gametes with the nutritional availability. Additionally, mature oocytes arrest both in meiosis I and meiosis II, and signaling and nutritional pathways have been shown to regulate their formation, and maintenance, such that despite long periods of arrest, the oocyte quality is assured and errors in chromosome segregation and varied cytoplasmic events are minimal.

Описание: This detailed volume contains embryo culture techniques that should inspire embryologists to consider comparative studies. The species included in this volume represent a broad range of taxa, whose embryos have vastly different culture requirements and developmental characteristics. Among the species described in this volume, significant differences exist in the rates of cell division, the timing of the maternal to embryonic transition, the relative lipid content of the cytoplasm, the sensitivity of the embryo to specific environmental ions, the preferred nutrients and associated metabolic pathways used by the embryo, the timing and mechanisms of early lineage specification, the presence or absence of embryonic diapause, and the time from fertilization to implantation. Written for the highly successful Methods in Molecular Biology series, chapters include introduction to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible protocols, and tips on troubleshooting and avoiding known pitfalls.
Authoritative and practical, Comparative Embryo Culture: Methods and Protocols serves as an ideal aid for scientists seeking another species whose embryos have evolved a unique mechanism that could provide a valuable, comparative context for experimentation.

Описание: The book considers signaling events from the zygote embryo through to the blastocyst with relevant data from embryonic stem (ES) cells, including dialogue with the extracellular environment and with the maternal tract during the implantation process. Application of the knowledge described to improve the success of human and animal assisted conception is considered where appropriate, but the focus is largely on fundamental rather than applied cell/molecular biology, as this is the area that has historically been neglected. While the general features of metabolism during preimplantation development are well established, especially in terms of nutrient requirements, uptake and fate, remarkably little is known about early embryo signaling events, intracellular or intercellular, between individual embryos in vitro or with the female reproductive tract in vivo. This contrasts with the wealth of information on cell signaling in somatic cells and tissues, as a glance at any textbook of biochemistry illustrates. This lack of information is such that our understanding of the molecular cell biology of early embryos -- a prerequisite to defining the mechanisms which regulate development at this critical stage of the life cycle -- is seriously incomplete. This volume is the first to address this issue by describing the current state of knowledge on cell signaling during mammalian early embryo development and highlighting priority areas for research.

Описание: This volume contains the Proceedings of the Serono Symposium on Pre- implantation Embryo Development, held in Newton, Massachusetts, in 1991. The idea for the symposium grew out of the 1989 Serono Symposium on Fertilization in Mammals* at which preimplantation development was the predominant suggestion for a follow-up topic.

Описание: It is a genuine honor and a privilege of distinction to provide the foreword for the proceedings that follow. This marvelous symposium reflects the unique qualities of its two principal sponsors. Whenever the Oregon Regional Primate Research Center is involved in matters of science, we see a consistent record of exceptional quality in both the work is unequaled and the people who do it. Likewise, Serono Symposia, USA in assuring a world-class speakers' forum, utterly without taint of commercial influence. The proceedings published herein are a remarkable testimony, not only to the contributors, but to Drs. Brenner, Wolf, Stouffer, and Burnett, who have shepherded its conception, presentation, and publication. Readers will notice immediately one of the universal strengths of this total composition; that is, the diversity of investigational interests among attendees. Presentations on the core topic of primate in vitro fertilization and embryo transfer were joined by specialist presentations in related areas, including human-assisted reproductive technologies, reproductive physiology of the great apes, sperm biology, implantation mechanisms, cryobiology, coculture systems for embryogenesis, micromanipulation technologies, and genetic diagnosis of heritable diseases in the pre embryo. Even though pro-fertility issues understandably dominated this forum, there was significant attention given to fertility-control research, especially the role of primates in this endeavor (more on this subject below)."
