Описание: This book will provide current understandings about two ubiquitously expressed metabotropic GPCRs, G-coupled purinoreceptor type 2 (P2Y) and Takeda G-protein-coupled bile acid receptor 5 (TGR5). G protein coupled receptors (GPCRs) are the largest family of proteins implicated in majority of cellular responses. The two receptor sub-families play a central role in many physiological functions as well as in many pathological conditions. This book offers up-to-date information on the physiological functions, signaling pathways and regulatory mechanisms of P2Y and TGR5 receptors. In addition, this book provides a comprehensive overview about the abnormalities of P2Y/TGR5 receptors and their contribution in the development and progression of pathological conditions. It also covers the currently available natural, chemical and pharmacological agents targeting these two receptor families and their therapeutic implications in P2Y and TGR5 associated disorders. This book is a valuable source for beginners and researchers to follow the rapidly progressing field of these two GPCR subfamily members.
Дополнительное описание: Module 1. Overview of G-Protein coupled receptor.- Module 2. TGR5 receptor.- Module 3. P2Y receptor.- Module 4. Therapeutically Targeting TGR5 & P2Y receptors.

Описание: It is the goal of The Metabotropic Glutamate Receptors to provide acomprehensive and forward-thinking review ofthe tremen- dous advances that have occurred in less than a decade of metabotropic glutamate receptors (mGluR) research. Virtually every areaof mGluR research is covered, including the molecular biology, pharmacology, anatomical distribution, and physiological and pathological roles of mGluRs. It is our intention that this volume not only summarize what is now known about the mGluRs, but also illuminate the areas in which there is the greatest need for focused research. Glutamic acid is an amino acid that has long been known to play several important metabolic roles in central and peripheral tissues and to be a component of several naturally occurring molecules. The first evidence that glutamate mayaIso serve as a neurotransmitter in the central nervous system (CNS) came in the late 1950s and early 1960s when glutamate and other acidic amino acids were found to induce behavioral convulsions when topically applied to the cortex and to excite a wide variety of central neurons. These findings spurred a massive research effort that quickly established glutamate as the pri- mary excitatory neurotransmitter in the vertebrate CNS. One of the most striking characteristics of glutamate that was quickly recognized was its ubiquitous role in serving as the neurotransmitter at the vast majority of excitatory synapses in the brain. It is now clear that most central neuronal circuits involve glutamatergic neurotransmission at some level.

Описание:

This book introduces readers to the latest advances in G protein-coupled receptor (GPCR) biology. It reviews our current understanding of the structural basis of ligand binding and allosteric mechanisms, following a decade of technological breakthroughs. Several examples of structure-based drug discovery are presented, together with the future challenges involved in designing better drugs that target GPCRs. In turn, the book illustrates the important concept of GPCR biased signaling in physiological contexts, and presents fluorescent- and light-based methodologies frequently used to measure GPCR signaling or to trace their dynamics in cells upon ligand activation.
Taken together, the chapters provide an essential overview and toolkit for new scientific investigators who plan to develop GPCR projects. All chapters were written by experts in their respective fields, and share valuable insights and powerful methodologies for the GPCR field.
