Описание: The calcified tissues have fundamental functions in the biology of organisms, not only because their strength, solidity, and elasticity permit movement and mechanical activities, and protect soft tissues against traumatic forces, but also on account of their role in mineral homeostasis. For this reason, extensive investigation in the last 30 years has provided much to explain the complex chemical and physical processes occurring in cells and matrices composing the skeleton, and their alterations in pathological conditions. The use of ultrastructural methods such as immunocytochemistry, scanning and transmission electron microscopy, cytoautoradiography, freeze/fracture etching, high voltage, etc.

has proven to be of great value when applied to cells and matrix components of bone and cartilage, in spite of the technical difficulties due to the hardness of these tissues. However, available information on this subject is disseminated in a variety of scientific and medical articles. This volume is an attempt to collect together the most significant data on the ultrastructure of cartilage and bone in normalcy and pathology.

Obviously, it cannot be a complete report of all these data, its principal aim being that of: a) giving a comprehensive statement of the results concerning the basic structures common to these tissues, especially collagen fibrils, noncollagenous proteins, and proteoglycans, and their relationships with the mineral substance (for which another volume of this series can also be consulted; see Ruggeri A. , Motta P. M.

Описание:

This textbook describes the biomechanics of bone, cartilage, tendons and ligaments. It is rigorous in its approach to the mechanical properties of the skeleton yet it does not neglect the biological properties of skeletal tissue or require mathematics beyond calculus. Time is taken to introduce basic mechanical and biological concepts, and the approaches used for some of the engineering analyses are purposefully limited. The book is an effective bridge between engineering, veterinary, biological and medical disciplines and will be welcomed by students and researchers in biomechanics, orthopedics, physical anthropology, zoology and veterinary science.

This book also:

Maximizes reader insights into the mechanical properties of bone, fatigue and fracture resistance of bone and mechanical adaptability of the skeleton

Illustrates synovial joint mechanics and mechanical properties of ligaments and tendons in an easy-to-understand way

Provides exercises at the end of each chapter

Описание: This volume focuseson the cell biology and physiology of skeletal muscle regeneration. This Bookis a collection of classic and cutting edge protocols optimized for mice, butin most cases adaptable to rat or other mammalian models, that will allow aninvestigator to develop and implement a research study on skeletal muscleregeneration. Chapters address the three major areas of study: provoking regeneration by inducing damage to muscle, analyzing theprogenitor cells of skeletal muscle, and quantifying overall muscle function. Subjects discussed include: inducing skeletal muscle injury byeccentric contraction; volumetric muscle loss; single myofiber isolation andculture; satellite cell transplantation; muscle clearing for whole mountimmunostaining; luciferase tracking of muscle stem cells; mitochondrialand mitophagy flux analysis; in vivo assessment of muscle contractility;force measurements on single isolated myofibers; and analysis of aerobicrespiration in intact skeletal muscle tissue by microplate respirometry. Written in the highlysuccessful Methods in Molecular Biology series format, chapters include introductions to each respective topic, lists of the necessarymaterials and reagents, step-by-step, readily reproducible laboratoryprotocols, and tips on troubleshooting and avoiding known pitfalls.Cutting edge andpractical, Skeletal Muscle Regeneration in the Mouse: Methods and Protocols is an essential laboratory reference for research inskeletal muscle growth, damage, repair, degeneration, and regenerative therapyin the mouse model system.

Описание: This book describes the diverse roles that growth factors and cytokines play in skeletal muscle. The extracellular environment has profound effects on the biology of skeletal muscle. The soluble portion of this environment includes a rich milieu of growth factors and cytokines which have been shown to regulate virtually all facets of the response of skeletal muscle to external stimuli, whether it be exercise induced metabolic shifts, remodeling in response to trauma or loading of the ongoing pathology associated with neuromuscular disease. The chapters included in this work illustrate growth factors that directly affect skeletal muscle cells and those which influence non-muscle cells that contribute to the biology of skeletal muscle as a whole tissue. The current state of the art, with the advent of systems biology, allows for the delineation of signaling networks which are regulated by suites of growth factors. This is in stark contrast to early more traditional studies, which only examined the effects of isolated growth factors on the activity of skeletal muscle precursor cells in tissue culture. The work presented in this volume ranges from reviewing and analyzing the roles of individual growth factors in detail, to the complex interplay of multiple soluble factors in the control of muscle functional, and dysfunctional states. The material covered in this volume will particularly suit readers from a range of research fields spanning general muscle biology and physiology, and those working on diseases and conditions affecting skeletal muscle both directly and indirectly.
