Описание: KENNETH A. FOON and ALTON C. MORGAN, JR. Passive immunotherapy using heteroantisera for the treatment of cancer in animals and humans has been studied for over 50 years. Attempts have been made to treat animal tumors with sera from immunized syngeneic, allogeneic, or xenogeneic animals. A number of studies of passive immunotherapy using heterologous antisera in humans have also been performed. These studies have generally been attempted in patients with large tumor burdens, and as would be expected, results have been transient at best. A wide variety of solid tumors as well as leukemias and lym- phomas have been treated with antisera raised in sheep, horses, rabbits, and goats. Problems such as anaphylaxis, serum sick- ness, and severe cytopenias have been encountered with these antisera. There are a number of potential mechanisms by which unconju- gated antibodies might be cytotoxic to tumor cells. Antibodies bound to the cell surface membrane of tumor cells may lead to cell lysis by complement-dependent or antibody-dependent cellu- lar cytotoxicity. Circulating tumor cells bound by antibody may be more susceptible to phagocytosis by the reticuloendothelial system. Antibody bound to the cell surface membrane of tumor cells may enhance immunogenicity of the tumor cell leading to activation of the host's immune system.

Описание: The concept of using bispecific antibodies for cancer therapy by retargeting immune effector cells was developed several years ago. Initial clinical studies were rather disappointing mainly due to low efficacy, severe side effects and the immunogenicity of the bispecific antibodies. The progress in antibody engineering finally led to the generation of new classes of bispecific antibodies lacking these obstacles. In addition, new applications were established, such as pre-targeting strategies in radioimmunotherapy and dual targeting approaches in order to improve binding, selectivity and efficacy. In this book, the different ways of generating bispecific antibodies are described, with emphasis on recombinant formats. The various applications of bispecific antibodies, e.g. in cellular cancer immunotherapy, radioimmunotherapy and pretargeting strategies are covered, and emerging applications such as dual targeting strategies, which involve the simultaneous inhibition of two targets, are addressed.

Описание: The development of monoclonal antibodies to human tumor associated antigens has greatly facilitated the application of immunohistochemical techniques to analyze surgically removed tissues. During the last few years this approach has been utilized by a progressively increasing number of investigators to analyze malignant cells. Although monoclonal antibodies to tumor associated antigens have not become yet routine reagents in immunopathology, they have provided new information which could not be obtained with conventional antisera or histochemical procedures. The following are representative examples. TUmor associated antigens have been identified which display a restricted distribution in normal tissues and therefore may represent useful markers for radio­ imaging and appropriate targets for immunotherapy. In spite of undetec­ table differences with conventional histopathological approaches hetero­ geneity has been found in the antigenic profile of tumor cells within a lesion, in autologous lesions removed from different anatomic sites from a given patient and in lesions removed from different patients. Phenotypes of tumor cells have been identified which correlate with the biology of tumor cells and with the clinical course of the disease. From a practical view point the use of monoclonal antibodies in immunopathology has enhanced interactions between pathologists and immunologists, as exemplified by the present book. Such interactions have contributed to the application of basic research to clinical problems. The chapter of this book discuss investigations performed with monoclonal antibodies to antigens expressed by various types of normal and malignant human cells.
