Описание: The interplay between tumors and their immunologic microenvironment is complex, difficult to decipher, but its understanding is of seminal importance for the development of novel prognostic markers and therapeutic strategies. The present review discusses tumor-immune interactions in several human cancers that illustrate various aspects of this complexity and proposes an integrated scheme of the impact of local immune reactions on clinical outcome. Current active immunotherapy trials have shown durable tumor regressions in a fraction of patients. However, clinical efficacy of current vaccines is limited, possibly because tumors skew the immune system by means of myeloid-derived suppressor cells, inflammatory type 2 T cells and regulatory T cells (Tregs), all of which prevent the generation of effector cells. To improve the clinical efficacy of cancer vaccines in patients with metastatic disease, we need to design novel and improved strategies that can boost adaptive immunity to cancer, help overcome Tregs and allow the breakdown of the immunosuppressive tumor microenvironment.

Описание: The focus of this book is a detailed discussion of a dual cancer vaccine (CV)-immune checkpoint inhibitor (ICI) mathematical model formulated as a system of partial differential equations (PDEs) defining the spatiotemporal distribution of cells and biochemicals during tumor growth.A computer implementation of the model is discussed in detail for the quantitative evaluation of CV-ICI therapy. The coding (programming) consists of a series of routines in R, a quality, open-source scientific computing system that is readily available from the internet. The routines are based on the method of lines (MOL), a general PDE algorithm that can be executed on modest computers within the basic R system. The reader can download and use the routines to confirm the model solutions reported in the book, then experiment with the model by varying the parameters and modifying/extending the equations, and even studying alternative models with the PDE methodology demonstrated by the CV-ICI model.Spatiotemporal Modeling of Cancer Immunotherapy: Partial Differential Equation Analysis in R facilitates the use of the model, and more generally, computer- based analysis of cancer immunotherapy mathematical models, as a step toward the development and quantitative evaluation of the immunotherapy approach to the treatment of cancer. To download the R routines, please visit: http://www.lehigh.edu/~wes1/ci_download

Описание: This timely book, published just as cancer immunotherapy comes of age, summarizes the rationale, present status, and future perspective for cancer immunotherapy. Included are explanations of the constitution of the immune system and immunocheckpoints, the mechanism of antigen presentation and recognition, valuable modalities, clinical trials and guidance, personalization, and biomarkers, all of which are essential for understanding the success of cancer immunotherapy. This innovative therapy has been investigated worldwide as the fourth line of cancer treatment after the standard treatments of surgery, chemotherapy, and radiotherapy. The progress in fundamental understanding of tumor immunology and the recent advances in clinical trials have opened new avenues with a cancer vaccine in 2010 and immunocheckpoint modulation in 2011, with their approval already granted in the United States. Today, there are no doubts, even among experts in cancer chemotherapy and radiotherapy, that the immune system plays a vital role in tumor eradication. Following American approval, many clinical trials of cancer immunotherapy are being conducted. With this book the reader will readily understand the paradigm shift in cancer treatment and will realize the importance of cancer immunotherapy. The great value of immunotherapy will be obvious, not only for tumor shrinkage but for prolonging patient survival.

Описание: This volume focuses on recent advances in understanding T cells as key players in antitumor immune responses, and as a result T cell-based immunotherapy is starting to transform the treatment of advanced cancers. However, despite recent successes, many patients with cancer fail to respond to these treatments. Defective migration of T cells into and within tumors is considered as an important resistance mechanism to cancer immunotherapy.

The volume includes three sections. The first section covers general knowledge about T cell trafficking during a normal immune response but also during tumor development. The second section provides an in-depth description of the different obstacles that prevent T cells from migrating and contacting tumor cells. The third section explores therapeutic strategies to improve trafficking of T cells into tumors and, thus, to enhance the effectiveness of cancer immunotherapy.

Описание:

Developing Costimulatory Molecules for Immunotherapy of Diseases highlights the novel concept of reverse costimulation and how it can be effectively exploited to develop immunotherapy using either humanized antibodies against CD80, CD86, and other costimulatory molecules or CD28 fusinogenic proteins in the treatment of diseases, including allergies, asthma, rheumatoid arthritis, multiple sclerosis, lupus nephritis, severe psoriasis, vulgaris tuberculosis, thopoid, transplantation therapeutic, cancer, and inflammation.

The text aims to provide the latest information on the complex roles and interactions within the CD28 and B7 costimulatory families, with the hope that targeting these families will yield new therapies for the treatment of inflammation, autoimmunity, transplantation, cancer, and other infectious diseases.
