Описание: This 8-chapter book is written by experts from China, Mexico, Portugal, United Kingdom and the United States of America. The book is focused on estrogen receptor structure, functions and clinical significance. Though we are unable to address all aspects of the estrogen receptors, we have analysed and discussed estrogen receptors or their isoforms in some major estrogen receptor-related disorders including cancers, endocrine diseases, respiratory diseases and obesity. The information provided by this book should aid our understanding the role of estrogen receptors in human diseases. This book will be of great value not only for clinicians interested in estrogen receptor-related diseases but also for basic scientists working in the field of estrogen receptors.

Описание:

Nuclear Receptors focuses on the structural analysis of nuclear receptors from the initial work using isolated protein domains to the more recent exciting developments investigating the conformational shape of full-length receptor complexes. The book also reviews the structure of key nuclear receptor co-regulatory proteins. It brings together, for the first time, a comprehensive review of nuclear receptor structure and the importance of receptor conformation underpinning allosteric regulation by different ligands (hormone, drugs, DNA response elements, protein-protein interactions) and receptor activity.

The nuclear receptor superfamily, including receptors for steroid hormones and non-steroid ligands, are pivotal to normal physiology, regulating processes as diverse as reproduction, metabolism, the immune system and brain development. The first members of the family were cloned over 25 years ago, which heralded in the idea of a superfamily of intracellular receptor proteins that bound small molecule ligands: classical steroid hormones, vitamins, fatty acids and other products of metabolism. These signals are then transmitted through multiprotein receptor-DNA complexes, leading to the regulation of target genes, often in a cell-selective manner. The cloning of the receptor cDNAs also ushered in an era of unparalleled analysis of the mechanisms of action of these ligand-activated transcription factors.

This volume examines a wide array of vital technologies for advancing our understanding of the receptor-mediated actions of estrogen. The protocols in this book range from standard methods and vital laboratory workhorses, such as receptor binding assays and western blot, to newer technologies such as RNAseq and proximity ligation assay. Chapters also discuss protocols from a broad range of tissue types to demonstrate the variety of estrogen receptor effects. Written in the highly successful Methods in Molecular Biology series format, chapters include introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and tips on troubleshooting and avoiding known pitfalls.

Cutting-edge and thorough, Estrogen Receptors: Methods and Protocols is a detailed and helpful resource for scientists who are intrigued by the many facets of estrogen.

The chapter 'Bioinformatics Analysis of Estrogen-Responsive Genes' is open access under a CC BY 4.0 license.

Описание: The increasing awareness on the varied consequences of hypogonadism in distinct organs and systems has supported the notion of estrogens as systemic agents. This observation is congruent with the variety of tissues affected by - trogens when used in hormone therapy formulations on hypogonadic women. Apart from the genital tract and the breast, recognized as traditional targets for estrogens, the skeleton, the vascular tree, or the central nervous system, are good examples of territories that have demonstrated sensitivity to estrogens. This evidence has created great interest, as shown by the great amount of lit- ature that has been produced on the bene?ts and risks associated with the use of estrogens. In parallel to the clinical interest, basic research has improved our kno- edge on the complexities involved in estrogen action at the molecular level. Together with effects mediated through speci?c receptors, a concept that has been the mainstay of the interpretation of estrogen action for years, there is enough evidence to hold the notion of receptor-independent effects. The substantial advances in modern technology applied to research have helped in enlightening the particulars of this versatile action of estrogens. This more detailed knowledge on the sophisticated mechanism of action of estrogens has nourished the emergence of multiple hypotheses speculating with the p- sibility of manipulating estrogen action. The notion that a widely extended regulatory system of cell function, as it is the estrogen receptor machinery, might be modulated at wish has arisen as an attractive, although still elusive postulate.

Описание: Estrogens are key modulators of brain regions that mediate learning and memory formation. This regulation has significant translational implications, as estrogens contribute to age-related memory decline and dementia, emotional disorders, addiction, and recovery from brain injury. Although the
importance of estrogens for memory formation has been well accepted within the behavioral neuroendocrinology community, it has yet to be fully appreciated by neuroscientists outside of the discipline.

Recent National Institutes of Health policies on the inclusion of sex as a biological variable in grant applications will force thousands of researchers to consider hormonal regulation of memory for the first time. However, the majority of these researchers are not trained endocrinologists, and no
previous monograph comprehensively encompasses the breadth of basic and clinical research on this subject. Thus, this book provides a vital resource for non-endocrinologists and endocrinologists alike.

Estrogens and Memory: Basic Research and Clinical Implications provides a compendium of cutting-edge basic and clinical research describing the ways in which estrogens regulate memory in a variety of species. Chapters are written by leading experts whose work is on the forefront of this exciting
field. Three fundamental discussions focus on: effects of estrogens on the hippocampus and other brain regions central to memory, effects of estrogens on memory and related cognitive processes throughout the lifespan, and translational implications of estrogenic regulation of memory for aging and
disease.

Описание: Since its ?rst description in 1942 in both serum and cerebrospinal ?uid, transthyretin (TTR) has had an eventful history, including changes in name from “prealbumin” to “thyroxine-binding prealbumin” to “transthyretin” as knowledge increased about its functions. TTR is synthesised in a wide range of tissues in humans and other eutherian mammals: the liver, choroid plexus (blood- cerebrospinal ?uid barrier), retinal pigment epithelium of the eye, pancreas, intestine and meninges. However, its sites of synthesis are more restricted in other vertebrates. This implies that the number of tissues synthesising TTR during vertebrate evolution has increased, and raises questions about the selection pressures governing TTR synthesis. TTR is most widely known as a distributor of thyroid hormones. In addition, TTR binds retinol-binding protein, which binds retinol. In this way, TTR is also involved with retinoid distribution. More recently, TTR has been demonstrated to bind a wide variety of endocrine disruptors including drugs, pollutants, industrial compounds, heavy metals, and some naturally occurring plant ?avonoids. These not only interfere with thyroid hormone delivery in the body, but also transport such endocrine disruptors into the brain, where they have the potential to accumulate.

Описание: There is a strong interest in transthyretin in connection with protein evolution, medical and clinical research. Transthyretin is a protein found in human blood and cerebrospinal fluid. It is directly involved in the transport of thyroid hormones, and indirectly in that of retinol. This monograph presents various developments and discoveries.
