Описание: ight junctions (TJs) are cell-ceil adhesion belts that encircle epithelial and endothelial cells at the limit between the apical and the lateral Tmembrane. These junctions are crucial for the establishment of separate compartments in multicellular organisms and for the exchange of substances between the internal milieu and the external environment. The perception of TJs has changed over the years. From being regarded as static paracellular seals, they have come to be perceived as dynamic structures that adjust their morphol- ogy and function in response to physiological, pharmacological and pathologi- cal challenges. The roles that TJs play in epithelial and endothelial cells has also widened, and nowadays this structure is regarded not only as a fence that limits within the membrane, the movement of proteins and lipids between the apical and basolateral membranes, or as a gate that regulates in a size and charge selec- tive manner the transit of ion and molecules through the paracellular pathway, but also as a structure integrated by molecules that participate in the control of cell proliferation. These observations highlight the importance of understand- ing TJ physiology in order to develop effective strategies for the treatment of pathological conditions such as cancer and autoimmune diseases. This broader perception of TJs is reflected in all the chapters of the book and has been attained thanks to the identification in recent years of a wide array of proteins that constitute TJs in epithelial and endothelial cells as well as in central nervous system myelin.

Описание:
A Laboratory Guide to the Tight Junction offers broad coverage of the unique methods required to investigate its characteristics. The methods are described in detail, including its biochemical and biophysical principles, step-by-step process, data analysis, troubleshooting, and optimization. The coverage includes various cell, tissue, and animal models.
Chapter 1 provides the foundations of cell biology of tight junction. Chapter 2 covers the Biochemical approaches for paracellular channels and is followed by chapter 3 providing the Biophysical approaches. Chapter 4 describes and discusses Histological approaches for tissue fixation and preparation. Chapter 5 discusses Light microscopy, while chapter 6 presents Electron microscopic approaches. Chapter 7 covers Transgenic manipulation in cell cultures, including DNA and siRNA, Mutagenesis, and viral infection. Chapter 8 covers transgenic manipulation in mice, including: Knockout, Knockin, siRNA knockdown, GFP/LacZ reporter, and overexpression. The final chapter discusses the future developments of new approaches for tight junction research.
Researchers and advanced students in bioscience working on topics of cell junction, ion channel and membrane protein will benefit from the described methods. Clinicians and pathologists interested in tissue barrier diseases will also benefit from the biochemical and biophysical characterization of tight junctions in organ systems, and their connection to human diseases.

Описание: As the first detailed overview of the subject, Tight Junctions brought together diverse perspectives from leading investigators to provide a multidisciplinary overview of the field. Since its publication in 1992, there has been an explosion of information about tight junctions, particularly in the area of molecular structure, cl and molecular biology, genetics and physiology. The field has advanced so fast and so far that it influences many areas of biology and now has practical implications for pharmacology of drug delivery and understanding a wide range of diseases altering epithelial permeability.

Описание: This volume explores the dynamic topic of tight junctions. The book focuses on tight junctions' role in sealing adjacent epithelial cells in a narrow band just beneath their apical surface. The book explains how tight junctions consist of a network of claudins and other proteins, and delves into how they hold cells together and form functional and protective barriers, regulating the passage of molecules and ions through the space between cells. The book opens with a discussion of the evolution of research on tight junctions, discussing a range of primary areas of growth, including the current knowledge on the particular physiological function of different claudins due to the development of an array of knock out mice. The chapters also explore in-depth studies of tight junctions in specific tissues (gut, lung and endothelia). This book offers a comprehensive understanding on post-translational modifications of occludin and its impact on tight junction function, as well as the molecular composition and function of tricellular tight junctions. This volume is particularly relevant to students and scientists in neuroscience, cell biology, physiology, cell differentiation, and cancer research.
