Описание: This book provides an updated account of tight junctions of different epithelia and endothelia and addresses their anatomy, biochemistry, physiology, synthesis, assembly, and modulation. It also discusses the relationship between molecular structure, function, regulation, posttranslational modifications of tight junction proteins and their role in gut microbiota maintenance, epithelial-mesenchymal transition, and intestinal barrier dysfunction-associated disorders important for the progression of human cancers. This book presents the pathological alterations of tight junctions at the blood-brain barrier that intensify or even initiate neurological dysfunction. Further, it reviews the significance of tight junctions and tight junction proteins in cancer development, infectious diseases, allergic reactions, and autoimmune disorders. Lastly, it highlights strategies for modulating tight junctions to enhance drug availability and organ deposition. This book is immensely useful for basic, translational, clinical, and interdisciplinary cancer researchers, practicing oncologists, and immunologists.
Дополнительное описание: Chapter 1. Inflammation and Cancer: Role of Tight Junctions.- Chapter 2. Regulation of tight junction by cadherin adhesion and its implication in inflammation and cancer.- Chapter 3. Tight junctions, epithelial-mesenchymal transition and cancer metastasis

Описание: This book overviews cancer immunity from broad scientific fields, based on the concept that cancer is a sort of by-product of infection, inflammation, and host immune response. The innate and acquired arms of the immune system mainly participate in tumor immune surveillance, and their activation is critically modulated by the situation of the tumor microenvironment. Many types of immune cells join the formation of the microenvironment. In particular, macrophages and dendritic cells enter the tumor mass to be main players in the inflammatory milieu of tumors. After introducing these topics, the book discusses immunotherapy for cancer patients as an outgrowth of this concept of infection and inflammation. With the contributions of leading scientists actively involved in the field of antitumor immunity study, this book encourages readers to understand the mechanism of general cancers based on inflammation and will facilitate prevention and the development of therapeutics for cancer.

Описание: This volume explores the dynamic topic of tight junctions. The book focuses on tight junctions' role in sealing adjacent epithelial cells in a narrow band just beneath their apical surface. The book explains how tight junctions consist of a network of claudins and other proteins, and delves into how they hold cells together and form functional and protective barriers, regulating the passage of molecules and ions through the space between cells. The book opens with a discussion of the evolution of research on tight junctions, discussing a range of primary areas of growth, including the current knowledge on the particular physiological function of different claudins due to the development of an array of knock out mice. The chapters also explore in-depth studies of tight junctions in specific tissues (gut, lung and endothelia). This book offers a comprehensive understanding on post-translational modifications of occludin and its impact on tight junction function, as well as the molecular composition and function of tricellular tight junctions. This volume is particularly relevant to students and scientists in neuroscience, cell biology, physiology, cell differentiation, and cancer research.

Описание: Neighboring cells are linked to each other by multimolecular complexes such as adherens junctions, desmosomes, and gap junctions. These complexes help maintain tissue integrity, act as barriers to permeability, reinforce cell polarity, and allow cells to communicate with each other.

Written and edited by experts in the field, this collection from Cold Spring Harbor Perspectives in Biology reviews our understanding of the organization, regulation, and dynamics of cell-cell junctions and the roles they play in morphogenesis, tissue homeostasis, and disease. The contributors examine the assembly and structure of different cell-cell adhesion systems, the plasticity of cell-cell junctions (e.g., during cell migration), and how the junctions act as hubs to sense and transduce various mechanical and chemical signals.

The authors also discuss the roles of cell-cell junctions in specific developmental and physiological processes, such as hearing, skeletal myogenesis, and neural circuit assembly, as well as in diseases such as cancer. This volume is therefore an indispensable reference for cell and developmental biologists, as well as anyone interested in understanding the roles of these complexes in human health and disease.

Описание: ight junctions (TJs) are cell-ceil adhesion belts that encircle epithelial and endothelial cells at the limit between the apical and the lateral Tmembrane. These junctions are crucial for the establishment of separate compartments in multicellular organisms and for the exchange of substances between the internal milieu and the external environment. The perception of TJs has changed over the years. From being regarded as static paracellular seals, they have come to be perceived as dynamic structures that adjust their morphol- ogy and function in response to physiological, pharmacological and pathologi- cal challenges. The roles that TJs play in epithelial and endothelial cells has also widened, and nowadays this structure is regarded not only as a fence that limits within the membrane, the movement of proteins and lipids between the apical and basolateral membranes, or as a gate that regulates in a size and charge selec- tive manner the transit of ion and molecules through the paracellular pathway, but also as a structure integrated by molecules that participate in the control of cell proliferation. These observations highlight the importance of understand- ing TJ physiology in order to develop effective strategies for the treatment of pathological conditions such as cancer and autoimmune diseases. This broader perception of TJs is reflected in all the chapters of the book and has been attained thanks to the identification in recent years of a wide array of proteins that constitute TJs in epithelial and endothelial cells as well as in central nervous system myelin.
