Описание: The work reported in this book represents an excellent example of how creative experimentation and technology development, complemented by computational data analysis, can yield important insights that further our understanding of biological entities from a systems perspective. The book describes how the study of a single RNA-binding protein and its interaction sites led to the development of the novel ‘protein occupancy profiling’ technology that for the first time captured the mRNA sequence space contacted by the ensemble of expressed RNA binders. Application of protein occupancy profiling to eukaryotic cells revealed that extensive sequence stretches in 3’ UTRs can be contacted by RBPs and that evolutionary conservation as well as negative selection act on protein-RNA contact sites, suggesting functional importance. Comparative analysis of the RBP-bound sequence space has the potential to unravel putative cis-acting RNA elements without a priori knowledge of the bound regulators. Here, Dr. Munschauer provides a comprehensive introduction to the field of post-transcriptional gene regulation, examines state-of-the-art technologies, and combines the conclusions from several journal articles into a coherent and logical story from the frontiers of systems-biology inspired life science. This thesis, submitted to the Department of Biology, Chemistry and Pharmacy at Freie Universit?t Berlin, was selected as outstanding work by the Berlin Institute for Medical Systems Biology at the Max-Delbrueck Center for Molecular Medicine, Germany.

Описание: Transcriptomics and proteomics, studying the profile of the expression of nucleic acids and proteins respectively, are increasingly applied to gain a mechanistic insight into a wide spectrum of investigation, and the use of expression profiling studies for the central nervous system and brain function aids in the understanding of neurodegenerative disorders and tumor development mechanisms. In Expression Profiling in Neuroscience, expert researchers provide a survey of the most commonly used approaches in the field and scan the different ways of studying the central nervous system/brain environment through expression profiling. The first part addresses the gene expression profiling of the brain at a large scale or to a specific cell type such as blood-brain barrier endothelium. Then, the second part describes the protein expression studies and the different technologies applied. Written for the popular Neuromethods series, chapters include corresponding background information, tested laboratory protocols, and step-by-step methods for reproducible laboratory experiments. Detailed and authoritative, Expression Profiling in Neuroscience presents the state-of-the-art techniques necessary to expand research further into this vital area of study.

Описание:

"Proteomic Profiling and Analytical Chemistry: The Crossroads, Second Edition" helps scientists without a strong background in analytical chemistry to understand principles of the multistep proteomic experiment necessary for its successful completion. It also helps researchers who do have an analytical chemistry background to break into the proteomics field. Highlighting points of junction between proteomics and analytical chemistry, this resource links experimental design with analytical measurements, data analysis, and quality control. This targeted point of view will help both biologists and chemists to better understand all components of a complex proteomic study.

The book provides detailed coverage of experimental aspects such as sample preparation, protein extraction and precipitation, gel electrophoresis, microarrays, dynamics of fluorescent dyes, and more. The key feature of this book is a direct link between multistep proteomic strategy and quality control routinely applied in analytical chemistry. This second edition features a new chapter on SWATH-MS, substantial updates to all chapters, including proteomic database search and analytical quantification, expanded discussion of post-hoc statistical tests, and additional content on validation in proteomics.
Covers the analytical consequences of protein and peptide modifications that may have a profound effect on how and what researchers actually measureIncludes practical examples illustrating the importance of problems in quantitation and validation of biomarkersHelps in designing and executing proteomic experiments with sound analytics
