Описание: This book provides a detailed account of the most recent developments, challenges and solutions to seamlessly advance and launch a lyophilized biologics or vaccine product, based on diverse modalities, ranging from antibodies (e.g., monoclonal, fused), complex biologics (e.g., antibody drug conjugate, PEGylated proteins), and vaccines (e.g., recombinant-protein based). The authors adeptly guide the reader through all crucial aspects, from biophysical and chemical stability considerations of proteins, analytical methods, advances in controlled ice nucleation and quality-by-design approaches, alternate drying technology, to latest regulatory, packaging and technology transfer considerations to develop a stable, safe and effective therapeutic protein, vaccine and biotechnology products. Lyophilized Biologics and Vaccines: Modality-Based Approaches is composed of four sections with a total of 17 chapters. It serves as a reference to all critical assessments and steps from early pre-formulation stages to product launch: * Provides recent understanding of heterogeneity of protein environment and selection of appropriate buffer for stabilization of lyophilized formulations * Details the latest developments in instrumental analysis and controlled ice nucleation technology * Explains in-depth lyophilized (or dehydrated) formulation strategies considering diverse modalities of biologics and vaccines, including plasmid DNA and lipid-based therapeutics * Details an exhaustive update on quality-by-design and process analytical technology approaches, illustrated superbly by case studies and FDA perspective * Provides the latest detailed account of alternate drying technologies including spray drying, bulk freeze-drying and crystallization, supported exceptionally by case studies * Provides a step-by-step guide through critical considerations during process scale-up, technology transfer, packaging and drug delivery device selection, for a successful lyophilization process validation, regulatory submission and product launch Chapters are written by one or more world-renowned leading authorities from academia, industry or regulatory agencies, whose expertise cover lyophilization of the diverse modalities of biopharmaceuticals. Their contributions are based on the exhaustive review of literature coupled with excellent hands-on experiences in laboratory or GMP setup, making this an exceptional guide to all stages of lyophilized or dehydrated product development.

Описание: The induction of antigen-specific immune responses after in vivo transfection with expression plasmids has triggered a revolution of vaccine research. After a first hype, evoked by the fascinating options of this method, clinical studies did not reach the ambitious aims and a phase of disillusion ensued. It became obvious that Gene vaccines displayed a weaker immunogenicity in humans than had been observed in the mouse models. Meanwhile these hurdles have been overcome and gene vaccines undergo a renaissance. The present book gives an update of the “world of naked gene vaccines”, namely DNA and RNA vaccines. Its content ranges from general mechanisms, inherent immunostimulatory properties and the vast potential to modulate immune responses, to recent successful clinical studies and approved veterinary gene vaccines. Beyond the state-of-the-art of genetic immunization, the reader will be stimulated with a chapter addressing “burning questions”.

Описание: This book offers an excellent introduction to the use of novel therapeutic vaccines for common diseases based on their ability to induce antibody production. While the role of vaccines in the treatment of infectious diseases and cancer is well known, vaccines have also recently been developed for a variety of other conditions, including Alzheimer’s disease, hypertension, diabetes, and spondyloarthritis. These therapeutic advances are fully and clearly documented by acknowledged experts in the field, who explain the relevant biology and highlight the challenges involved in deploying this treatment approach effectively and safely. In addition, recent progress in the construction and delivery of DNA vaccines is documented, and the process of developing new peptide vaccines is explored in depth. While the book will be particularly valuable for researchers and scholars interested in immunotherapy, it will also appeal to clinicians seeking effective new medicines to treat patients suffering from chronic diseases.

Описание: Why another book about vaccines? There are already a few extremely well-written medical textbooks that provide comprehensive, state-of-the-art technical reviews regarding vaccine science. Additionally, in the past decade alone, a number of engrossing, provocative books have been published on various related issues ra- ing from vaccines against specific diseases to vaccine safety and policy. Yet there remains a significant gap in the literature – the history of vaccines. Vaccines: A Biography seeks to fill a void in the extant literature by focusing on the history of vaccines and in so doing, recounts the social, cultural, and scientific history of vaccines; it places them within their natural, historical context. The book traces the lineage – the “biography” – of individual vaccines, originating with deeply rooted medical problems and evolving to an eventual conclusion. Nonetheless, these are not “biographies” in the traditional sense; they do not trace an individual’s growth and development. Instead, they follow an idea as it is conceived and dev- oped, through the contributions of many. These are epic stories of discovery, of risk-takers, of individuals advancing medical science, in the words of the famous physical scientist Isaac Newton, “by standing on the shoulders of giants. ” One grant reviewer described the book’s concept as “triumphalist”; although meant as an indictment, this is only partially inaccurate.
